1) Applications of the FLUOVIEW FV3000

  • High-Sensitivity Spectral Multi-Channel Imaging. FV3000 is equipped with High-Sensitivity Spectral Detector (HSD) with GaAsP Photomultiplier Tubes (PMTs) for spectral detection and up to 16 channel spectral unmixing.
  • Macro to Micro Imaging with Olympus Super Resolution. FV3000 supports quick macro to micro imaging from 1.25x to 150x. Olympus Super Resolution system (FV-OSR) enables lateral (xy) resolution of approximately 120 nm in up to 4 channels. Addition of Constrained Iterative Deconvolution allows to simultaneously improve axial (z) resolution.
  • High-Speed Confocal Imaging (30-438 frames per second). FV3000 is equipped with two scanners: galvo scanner for imaging larger field of view in high resolution and resonance scanner for very fast imaging (512x512 pixels at 30 fps to 512x32 pixels at 438 fps).
  • FRET (Förster Resonance Energy Transfer). FV3000 is suitable for imaging multiple FRET pairs and is equipped with analysis software for FRET experiments.
  • FRAP (Fluorescence Recovery After Photobleaching). FV3000 contains analysis package for fitting experimental FRAP data.
  • Accurate Multiposition 3D Time Lapse Imaging. ZDC system in FV3000 precludes focal drift in long-term experiments.

 

2) FV3000 Instrument specifications

Confocal laser scanning microscope Olympus FLUOVIEW FV3000 is an inverted fully motorized IX83 microscope equipped with:

Six lasers (all Coherent OBIS Solid-state lasers):

  • 405 nm (50 mW)
  • 445 nm (75 mW)
  • 488 nm (20 mW)
  • 514 nm (40 mW)
  • 561 nm (20 mW)
  • 640 nm (40 mW)

The selection of the laser line is controlled via an Acousto Optical Tunable Filter (AOTF). The use of an AOTF allows excitation intensity balancing within an image, flexible area bleaching e.g. for FRAP/Photo-activation experiments and line sequential scanning to avoid cross-talk between channels (multi-tracking).
